    Effects of Plantain (Plantago lanceolata L.) Metabolites Aucubin, Acteoside, and Catalpol on Methane Emissions In Vitro

    Plantain (PL) contains plant secondary metabolites (PSM), such as acteoside, aucubin, and catalpol, known for their bioactive properties. While acteoside and aucubin have been linked to reducing nitrogen losses in grazed pastures, their effects on enteric methane (CH4) emissions remain unexplored. Three in vitro batch culture experiments were conducted to assess the effects of PSM on rumen fermentation, using PL pastures with varying PSM concentrations, purified PSM compounds, and/or their combinations added to ryegrass (Lolium perenne, RG), which does not contain these PSM. Aucubin addition to RG extended the time to reach halftime for gas production (GP) and CH4 by 15-20% due to its antimicrobial effects. Acteoside, alone or with aucubin, promoted propionate production, an alternative hydrogen sink, which reduced the acetate to propionate ratio, increased GP by up to 13%, and decreased CH4 proportion in gas by 5-15%. Aucubin reduced ruminal net ammonia (NH3) production by up to 46%, with a similar reduction observed when combined with acteoside. This study highlights the potential of PSM to mitigate CH4 emissions and reduce nitrogen losses from dairy cows, warranting in vivo evaluation of PSM and targeted breeding of PL pastures with increased PSM content.

    Teat and udder morphology and pathology of New Zealand dairy ewes.

    AIMS: To describe the morphology and prevalence of teat and udder pathology of New Zealand dairy ewes. METHODS: A repeated cross-sectional study was conducted on 20 commercial New Zealand dairy sheep farms over the 2022-2023 season. Approximately 15 randomly selected ewes were examined on each farm in early, mid, and late lactation. Four udder morphology measures were scored on a 5-point scale: depth (1 = pendulous, 5 = compact udder), suspension (ratio of width at abdominal attachment to height, 1 = low, 5 = high), gland separation (1 = zero, 5 = maximum separation), and teat placement (1 = distally located, vertically oriented, 5 = laterally located, horizontally oriented teats). Teat length and width were measured, and the presence of supernumerary teats, asymmetry, visible teat/udder inflammation, and lesions were recorded. Teats and glands were palpated for consistency, gland-nodules, teat canal thickening, and patency, and teat ends scored for hyperkeratosis. Teat and gland-level pathological variables were converted to ewe-level scores. The presence of involuted, non- or minimally lactating glands was recorded. Data were analysed using descriptive statistics. RESULTS: Across the three visits, 893 observations were made on 882 unique ewes. Mean teat length and width were 27.5 (95% CI = 27.2-27.7) and 15.8 (95% CI = 15.7-15.9) mm. Udder depth, separation, suspension, and teat placement had modal scores of 4, 3, 3, and 3 respectively, and varied across visits and age groups. Asymmetrical udder prevalence was 39% (95% CI = 35.5-42%) overall and increased with age (27% for 1-year-olds, 38% for two-year-olds, 43% for mixed age ewes). Supernumerary teats were observed in 15% (95% CI = 13-17.9%) of ewes. There was between-farm variation in all udder morphology variables. Ewe-level prevalences of teat end hyperkeratosis (any degree of severity), gland/teat inflammation, lesions, palpable defects, and involuted glands were all < 6%. CONCLUSIONS: Morphological observations resembled those from overseas dairy sheep. Teat dimensions, udder depth, separation, suspension, teat placement, and presence of supernumerary teats varied between farms. Udder depth, separation, and suspension scores decreased with age, while teat placement score and the prevalence of asymmetry increased with age. Teat and udder pathology were rare. CLINICAL RELEVANCE: This is the first systematic study of teat and udder morphology and pathology in New Zealand dairy ewes. The data will be useful for farmers and industry partners, providing a comparison with their own flocks, identifying areas for improvement, and informing and contrasting with future studies.

    Effectiveness of dairy products to protect against cognitive decline in later life: a narrative review

    As the world's population ages the prevalence of age-related health concerns is increasing, including neurodegeneration disorders such as mild cognitive impairment, vascular dementia and Alzheimer's disease. Diet is a key modifiable risk factor for the development of neurodegeneration, likely due to gut-brain axis interactions related to neuroinflammation. Analyses of dietary patterns identified dairy as being part of a cognitively healthy diet; however, its contribution to cognitive outcomes is difficult to discern. This narrative review evaluates the literature to determine whether there is sufficient evidence that the consumption of dairy products helps to maintain cognitive function in later life. A search using the terms (dairy OR milk OR cheese OR yogurt OR yogurt) AND ("mild cognitive impairment" OR dementia OR "Alzheimer's disease") identified 796 articles. After screening and sorting, 23 observational studies and 6 intervention studies were identified. The results of the observational studies implied that the relationship between total dairy consumption and cognitive outcomes is inverse U-shaped, with moderate consumption (1-2 servings per day) being the most beneficial. The analysis of the intake of different types of dairy products indicated that fermented products, particularly cheese, were most likely responsible for the observed benefits. The experimental studies all used dairy-derived peptides produced during fermentation as the dietary intervention, and the results indicated that these could be an effective treatment for early-stage cognitive impairment. Further experimental studies with whole dairy products, particularly fermented dairy, are needed to determine whether the regular consumption of these foods should be recommended to maximize the likelihood of healthy cognitive aging.

    The impact of heat treatment of bovine milk on gastric emptying and nutrient appearance in peripheral circulation in healthy females: a randomized controlled trial comparing pasteurized and ultra-high temperature milk

    BACKGROUND: Heat treatments of dairy, including pasteurization and ultra-high temperature (UHT) processing, alter milk macromolecular structures, and ultimately affect digestion. In vitro, animal, and human studies show faster nutrient release or circulating appearance after consuming UHT milk (UHT-M) compared with pasteurized milk (PAST-M), with a faster gastric emptying (GE) rate proposed as a possible mechanism. OBJECTIVES: To investigate the impact of milk heat treatment on GE as a mechanism of faster nutrient appearance in blood. We hypothesized that GE and circulating nutrient delivery following consumption would be faster for UHT-M than PAST-M. METHODS: In this double-blind randomized controlled cross-over trial, healthy female (n = 20; 27.3 ± 1.4 y, mean ± SD) habitual dairy consumers, consumed 500 mL of either homogenized bovine UHT-M or PAST-M (1340 compared with 1320 kJ). Gastric content volume (GCV) emptying half-time (T50) was assessed over 3 h by magnetic resonance imaging subjective digestive symptoms, plasma amino acid, lipid and B vitamin concentrations, and gastric myoelectrical activity were measured over 5 h. RESULTS: Although GCV T50 did not differ (102 ± 7 min compared with 89 ± 8 min, mean ± SEM, UHT-M and PAST-M, respectively; P = 0.051), GCV time to emptying 25% of the volume was 31% longer following UHT-M compared with PAST-M (42 ± 2 compared with 32 ± 4 min, P = 0.004). Although GCV remained larger for a longer duration following UHT-M (treatment × time interaction, P = 0.002), plasma essential amino acid AUC was greater following UHT-M than PAST-M (55,324 ± 3809 compared with 36,598 ± 5673 μmol·min·L-1, P = 0.006). Heat treatment did not impact gastric myoelectrical activity, plasma appetite hormone markers or subjective appetite scores. CONCLUSIONS: Contrary to expectations, GE was slower with UHT-M, yet, as anticipated, aminoacidemia was greater. The larger GCV following UHT-M suggests that gastric volume may poorly predict circulating nutrient appearance from complex food matrices. Dairy heat treatment may be an effective tool to modify nutrient release by impacting digestion kinetics. CLINICAL TRIAL REGISTRY: www.anzctr.org.au (ACTRN12620000172909).

    Pasture Performance: Perspectives on Plant Persistence and Renewal in New Zealand Dairy Systems

    Pasture systems dominate the landscape of Aotearoa, New Zealand, and are an integral component of sustainable and resilient livestock production. Predicting the response, performance, and dynamics of pasture species and adapting management practices is key to the long-term economic and environmental sustainability and resilience of the agricultural sector. However, there is limited information on the long-term productivity, performance, and persistence of forage cultivars and species for pasture production systems, particularly when linked to grazing and animal performance. Here, we sought to reduce scientific uncertainty, inform modelling efforts, and contribute to a predictive framework for understanding pasture performance, persistence, and renewal. Inter-annual pasture renewal (direct drilling and cultivation) rates vary by region and year, reflecting both opportunity and problem-based drivers, with the highest pasture renewal rates in Waikato and Canterbury on the North and South Island, respectively.

    Novel hyperthermoacidic archaeal enzymes for removal of thermophilic biofilms from stainless steel

    AIMS: To test the efficacy of novel hot/acid hyperthermoacidic enzyme treatments on the removal of thermophilic spore-forming biofilms from stainless steel surfaces. METHODS AND RESULTS: The present study measured the efficacy of hyperthermoacidic enzymes (protease, amylase, and endoglucanase) that are optimally active at low pH (≈3.0) and high temperatures (≈80°C) at removing thermophilic bacilli biofilms from stainless steel (SS) surfaces. Plate counts, spore counts, impedance microbiology, as well as epifluorescence microscopy, and scanning electron microscopy (SEM) were used to evaluate the cleaning and sanitation of biofilms grown in a continuous flow biofilm reactor. Previously unavailable hyperthermoacidic amylase, protease, and the combination of amylase and protease were tested on Anoxybacillus flavithermus and Bacillus licheniformis, and endoglucanase was tested on Geobacillus stearothermophilus. In all cases, the heated acidic enzymatic treatments significantly reduced biofilm cells and their sheltering extracellular polymeric substances (EPS). CONCLUSIONS: Hyperthermoacidic enzymes and the associated heated acid conditions are effective at removing biofilms of thermophilic bacteria from SS surfaces that contaminate dairy plants.

    Extracellular Polysaccharide Extraction from Streptococcus thermophilus in Fermented Milk.

    Lactic acid bacteria such as Streptococcus thermophilus are known to produce extracellular polysaccharide (EPS) in fermented foods that enhance the creaminess and mouthfeel of the product, such as yogurt. Strains producing larger amounts of EPS are highly sought-after, and therefore, robust and accurate quantification methodologies are important. This study found that two commonly used methodologies significantly underestimated the amount of EPS produced as measured using a milk matrix. To this end, a proteolytic step was implemented prior to EPS extraction (Method C). An initial proteolytic step using xanthan gum-spiked milk significantly increased recovery yield to 64%, compared to 27.8% for Method A and 34.3% for Method B. Method C showed no improvement when assessed using a chemically defined medium. Method C was further validated using three strains of S. thermophilus with varying EPS-production capabilities (STLOW, STMID, STHIGH). Overall, Method C demonstrated significant improvements in the EPS extraction yield for all three S. thermophilus strains in fermented milk. On average, Method C improved isolation yield by ∼3- to 6-fold compared with Method A and by ∼2- to 3-fold compared with method B. There were no significant differences between samples when they were grown in a chemically defined medium, highlighting the importance of a proteolytic step specifically for fermented milk samples. In commercial applications, accurate quantification of EPS-production is an important aspect when finding new strains. IMPORTANCE Extracellular polysaccharide (EPS) production by milk-fermenting microorganisms is a highly sought-after trait in improving the perceived thickness, creaminess, and mouthfeel of yogurt. Streptococcus thermophilus are commonly isolated and their EPS production is quantified in the search for higher-producing strains. In this study, we demonstrated that two commonly used methods for isolating EPS from milk samples significantly underestimated the true amount of EPS present. We demonstrated that the addition of a proteolytic step prior to EPS extraction isolated over 2-fold more EPS than identical samples processed using the traditional protocols. We further validated this method in fermented milk samples from three strains of S. thermophilus that included a low-, mid-, and high-EPS producing strain. Again, we showed significant improvements in EPS isolation using a proteolytic step. In the search for new S. thermophilus strains with enhanced EPS production, accurate quantification in an optimal medium is essential.

    The characterisation of antimicrobial resistant Escherichia coli from dairy calves.

    Introduction. Dairy calves, particularly pre-weaned calves have been identified as a common source of multidrug resistant (MDR) Escherichia coli. Gap statement. E. coli strains isolated from dairy calves and the location of their resistance genes (plasmid or chromosomal) have not been well characterised. Aim. To characterise the phenotypic and genotypic features as well as the population structure of antimicrobial-resistant E. coli isolated from calves located on dairy farms that feed waste-milk to their replacement calves. Methodology. Recto-anal swab enrichments from 40 dairy calves (≤ 14 days old) located on four dairy farms were examined for tetracycline, streptomycin, ciprofloxacin, and third-generation cephalosporin resistant E. coli. Whole genome sequencing was performed using both short- and long-read technologies on selected antimicrobial resistant E. coli. Results. Fifty-eight percent (23/40) of calves harboured antimicrobial resistant E. coli: 43 % (17/40) harboured tetracycline resistant, and 23 % (9/40) harboured chromosomal mediated AmpC producing E. coli. Whole genome sequencing of 27 isolates revealed five sequence types, with ST88 being the dominant ST (17/27, 63 % of the sequenced isolates) followed by ST1308 (3/27, 11 %), along with the extraintestinal pathogenic E. coli lineages ST69 (3/27, 11 %), ST10 (2/27, 7 %), and ST58 (2/27, 7 %). Additionally, 16 isolates were MDR, harbouring additional resistance genes that were not tested phenotypically. Oxford Nanopore long-read sequencing technologies enabled the location of multiple resistant gene cassettes in IncF plasmids to be determined. Conclusion. Our study identified a high incidence of tetracycline and streptomycin-resistant E. coli in dairy calves, and highlighted the presence of multidrug-resistant strains, emphasising the need for further investigation into potential associations with farm management practices.

    Service Sire Effects on Body Condition Score, Milk Production, and Rebreeding of Mixed-Aged Dairy Cows Bred to Angus and Hereford Bulls—A Case Study in New Zealand

    Beef-breed bulls are used in dairy herds to produce a calf of greater value for finishing than calves sired by dairy bulls. There is limited research about which beef-breed bulls are most appropriate, and whether any negative impact on cow performance in terms of milk production and rebreeding should be considered. The aim of this case study was to compare the body condition score, milk production, and rebreeding performance of mixed-aged dairy cows bred to a selection of Angus and Hereford beef-breed bulls. Body condition score, post-calving live weight, milk production, rebreeding performance, and survival of 952 mixed-aged dairy cows artificially bred to 65 Angus and Hereford bulls were compared. There was no effect of service sire on post-calving live weight, days in milk, milk production, or inter-calving intervals of mixed-aged cows. Service sire had an effect on the calving day due to differences in gestation length (p < 0.001), although this did not translate into an effect on days in milk. A longer gestation length negatively influenced pregnancy rates, and greater birth weight of the calf negatively influenced survival to rebreeding (p < 0.05), indicating the potential for an effect of service sire. Selection of beef-breed service sires for dairy cows should include emphasis on lighter calves and shorter gestation lengths. The general absence of the service sire effect on the parameters measured in this study indicated that any of the service sires used in this experiment would be appropriate for use over dairy cows.

    The Effect of Lameness on Milk Production of Dairy Goats

    Lameness on dairy goat farms is a welfare concern and could negatively affect milk production. This study's objective was to evaluate the effects of clinical lameness on the daily milk production of dairy goats. Between July 2019 and June 2020, 11,847 test-day records were collected from 3145 goats on three farms in New Zealand. Locomotion scoring of goats used a five-point scoring system (0 to 4). The dataset was split into two groups by lactation type, where goats were classified as being in seasonal lactation (≤305 days in milk) or extended lactation (>305 days in milk). A linear mixed model was used to analyze datasets using milk characteristics as the dependent variables. Severely lame goats (score 4) in seasonal and extended lactation produced 7.05% and 8.67% less milk than goats not lame, respectively. When the prevalence of severe lameness is between 5 and 20% of the herd, the estimated average daily milk income lost was between NZD 19.5 and 104 per day. This study established the negative impact of lameness on milk production and annual income in dairy goats on three farms.
